PRODUCTION OF ACETOBACTER XYLINUM MICROBIAL
STARTER WITH LIME AND FOOD VINEGAR ADDITIONAL

ABSTRACT
Acetobacter xylinum are obligate aerobic bacteria that metabolize mainly glucose which is used in
the synthesis of cellulose. The floating matrix will allow the cells to rise to the surface of the media
where oxygen is abundant. The purpose of this study was to determine the addition of different acids
to A. xylinum starter. The method used to grow starter A. Xylinum consisted of two different
treatments, namely the addition of lime and 5% Food Vinegar. This research was tested
descriptively. Preparation of microbial starter Acetobacter xylinum using the addition of lime juice
gives faster changes every time compared to the 5% Food Vinegar treatment. In the fermentation
with the addition of white coating lime and color changes occurred on the second day, while the 5%
Food Vinegar treatment occurred on the sixth day.

1. INTRODUCTION
According to[1], The definition of a starter is a
number of additional ingredients that can be used
during the initial phase of the fermentation
process. Starter is a process that is intended to
grow certain microbes in a substrate or medium
that can make these microbes grow in a certain
purpose and process. The process of growing
bacteria using this starter method has conditions
that must be known such as the medium where it
must be clean, sterile, superior, the materials used
are in accordance with procedures, stable during
the process, and also not harmful pathogens.
According to [2] said that, the conditions for
making a starter are safe, and can also withstand
harmful bacteria in the process of making a starter
Microorganisms in the starter manufacturing
process will grow and reproduce simultaneously
and are active. This growing microbe will change
the material in the starter process into the
expected material [3]. Environmental factors and
nutrient content in the process of making this
starter will affect the growth rate of microbes. The
fermentation process will run if it uses the
benefits of glucose as an energy source. The
nutrients in the substrate or medium will decrease
as the fermentation process progresses and the
duration of the process. The large number of
microbes that grow and reproduce will result in
competition and will end in the death phase [1].
Acetobacter xylinum microbe is one of the
microbes that will often be encountered in the
field of fisheries. This microbe has benefits such
as making nata which is produced from solulose
fermented by Acetobacter xylinum. Acetobacter
xylinum bacteria can produce extracellular
enzymes that can polymerize glucose into
thousands of fiber or cellulose homopolymer
chains. There is an easy way to get specific and
structured Acetobacter xylinum microbes with
bacterial culture.
There are various types of fermentation, one of
which is starter fermentation. According to[4]
explained that, starter fermentation is a
fermentation that is processed in foodstuffs with
the addition of microorganisms that will play an
active role in changing the fermented material
into the expected product. These microorganisms
will multiply. In this case, we can use these
microorganisms as a role in production. The role
that is utilized in this microorganism is the
metabolic activity of the microorganism.
