AMR is one of the causes of global health problems that we must be aware. Raw milk is easily contaminated with bacteria, including E. coli. An Antimicrobial often used in cattle treatment is penicillin, that can kill bacteria by preventing the formation of cell wall proteins. This study aimed to detect the presence and prevalence of E. coli in raw milk and test its resistance to penicillin. Seventy fve raw milk samples were randomly selected from 6 groups of dairy cattle and collected by each feld ofcer. Identifcation of the presence of E. coli refers to the Indonesian National Standard (SNI) number 01-2897-2008 concerning the method of testing for microbial contamination in milk, meat, and eggs. Bacterial isolates were identifed as E. coli further to be tested for resistance to penicillin using the Kirby-Bauer disk difusion method with standard determination based on the Clinical and Laboratory Standards Institute (CLSI). Structured interviews were conducted with 75 farmers whose milk samples were taken. The data obtained were analyzed descriptively. The identifcation results showed that E. coli were detected in 24/75 isolates (32%). Resistance test results showed 23/24 E. coli isolates (96%) resistance to penicillin. The conclusion from this study is that E. coli, which is isolated from raw milk in the KPSCU, is resistant to penicillin, so it is necessary to practice of hygiene sanitation, monitoring antibiotic use, increase education for farmers and animal health workers on the use of antimicrobials in the area. Keywords: antibiotic resistance; Escherichia coli; milk; penicillin; questionnare Abstrak Resistansi antibiotik atau sering disebut sebagai pandemi senyap, merupakan satu diantara penyebab masalah kesehatan global yang harus diwaspadai. Susu segar merupakan media pembawa yang mudah terkontaminasi bakteri, salah satunya ialah bakteri E. coli. Antibiotik yang sering dipakai dalam pengobatan terhadap sapi adalah penisilin yaitu antibiotik golongan betalaktam yang memiliki kemampuan membunuh bakteri dengan mencegah pembentukan protein dinding sel. Tujuan penelitian ini adalah untuk mendeteksi keberadaan dan prevalensi E. coli pada susu segar, serta menguji resistansinya terhadap antibiotik penisilin. Sebanyak 75 sampel susu kandang dipilih secara acak, berasal dari 6 kelompok ternak sapi perah dan dikumpulkan oleh masing-masing petugas lapang. Uji identifkasi keberadaan E. coli mengacu kepada Standar Nasional Indonesia (SNI) nomor 01-2897-2008 tentang metode pengujian cemaran mikroba pada susu, daging dan telur.
