Lima 5 Resumo Objetivo: Rever de forma sistemática as substân- cias adjuvantes mais estudadas, utilizadas e as novas substâncias; com o intuito de atualizar os conceitos de sua ação por atuarem, juntamente com o antígeno, no sistema imunológico e seu futuro uso em vacinas hu- manas ou animais. Métodos: Foi realizada revisão sistemática em li- vros e da literatura publicada no Medline ® a partir do ano de 1969. Foram selecionados artigos que abordas- sem o tema “adjuvantes” nos seguintes aspectos: Res- posta inflamatória, resposta imune celular, resposta imune humoral, seqüelas, proteção. Resultados: Foram selecionados artigos originais ou capítulos de livros abordando este tema. Observou- se haver diferenças na capacidade de indução infla- matória entre os diferentes adjuvantes. Estas diferen- ças podem resultar em padrões de resposta imune ce- lular e humoral diferenciados, de acordo com a célula mais estimulada. Assim, adjuvantes indutores de mi- gração predominantemente mononuclear parecem fa- cilitar respostas do padrão celular e humoral. Respos- tas mais neutrofílicas podem resultar em maiores efei- tos colaterais locais e/ou sistêmicos e também podem ser associados à redução da disponibilidade anti- gênica para o sistema imunológico. Conclusões: O sistema imunológico animal ou hu- mano pode se comportar de maneira diferenciada frente aos diferentes tipos de substâncias adjuvantes. Os mesmos mecanismos usados pelo organismo para proteção contra infecções podem, em situações parti- culares, serem causadores de doenças, por isso a im- portância do estudo individual de cada adjuvante a ser Abstract: Objective: To systematically review the adjuvants used, to study the new substances with the aim of up- date the concepts of the use of these substances, which act together with the antigen, when in contact with the immunological system, and their use for human or animal vaccine. Methods: A literature systematic review was ac- complished based on Medline data bank starting from 1969. Original articles on adjuvants discussing in- flammatory response, cellular immune response, hu- moral immune response, side-effects and protection aspects were selected. Results: Original articles or book chapters were selected. We could observe that there are differences in the inflammatory capacity among the different ad- juvants. These differences can result in some patterns of cellular and humoral immune responses, in agree- ment with cell which is more stimulated. Therefore, adjuvants which induce mononuclear activation seem to facilitate cellular and humoral response. A more granulocytic response can result in more local or sys- temic side-effects and can also be associated with a reduction of antigen capacity for the immune system stimuli. Conclusion: The animal or human immunological system behaves different when challenged with some adjuvants. Once the same mechanisms used by the or- ganism for protection against infections can induce di- seases, there is an important aspect in studying each adjuvant to be used in a particular situation.
