TTh1)cellsinthepancreaticisletsplayapivotalroleinthepro-
gressionofdiabetesinnon-obesediabeticNOD)mice.Wede-
monstrate in the present report that a butanol fraction of B.
pilosa suppressedthedevelopmentofdiabetes,helpedmaintain
levelsofbloodsugarandinsulininNODmiceinadose-depen-
dentmannerandelevatedtheserumIgElevelsregulatedbyTh2
cytokinesinNODmice.Moreover,thebutanolfractioninhibited
thedifferentiationofnaïvehelperTTh0)cellsintoTh1cellsbut
enhancedtheirtransitionintotypeIIhelperTTh2)cellsusing
an in vitro Tcelldifferentiationassay.Twopolyacetyleniccom-
pounds, 2-b-D-glucopyranosyloxy-1-hydroxy-5E)-tridecene-7,9,11-
triyne and 3-b-D-glucopyranosyloxy-1-hydroxy-6E)-tetradecene-
8,10,12-triyne, identified from the butanol fraction also prevented
theonsetofdiabeteslikethebutanolfraction.Thelattercompound
showedastrongeractivityforTcelldifferentiationthantheformer.
Insummary,thebutanolfractionof B. pilosa anditspolyacetylenes
canpreventdiabetesplausiblyviasuppressingthedifferentiationof
Th0 cells into Th1 cells and promoting that of Th0 cells into Th2
cells.
