Penelitian menggunakan metode eksperimental menggunakan Rancangan Acak Kelompok (RAK) yang terdiri atas lima blok. Sebagai blok adalah periode pengambilan cairan rumen dari tiga ekor sapi potong yang diambil dari Rumah Potong Hewan (RPH) Mersi, Purwokerto. Perlakuan yang diuji adalah level ekstrak daun waru (Hibiscus tiliaceus) dalam konsentrat (mg/kg BK); 0 ppm, 50 ppm, 100 ppm, dan 150 ppm berturut-turut untuk RІ, RЇ, RЈ, daŶ RЉ. IŵbaŶgaŶ bahaŶ keriŶg pakaŶ koŵplit yaŶg diguŶakaŶ terdiri atas 45% jerami padi amoniasi dan 55% konsentrat. Peubah yang diukur adalah populasi protozoa dan kecernaan bahan kering (KBK). Analisis variansi menunjukkan bahwa pemberian ekstrak daun waru berpengaruh nyata (P < 0,05) terhadap populasi protozoa namun tidak berpengaruh (P > 0,05) terhadap KBK. Peningkatan konsentrasi daun waru (ppm) menurunkan populasi protozoa secara kuadrater (P < 0,05) dengan populasi terendah dicapai pada level 104,6 ppm. Rataan populasi protozoa yaitu 819200±123179.5 ; 678000±208734.3 ; 580000±198494.3 ; 648000±240977.2 (per ml cairan rumen) dan KBK yaitu 37.78±2.32 ; 36.49±2.40 ; 36.33±2.39 ; 45.68±19.07 (%) untuk berturut-turut RІ, RЇ, RЈ, daŶ RЉ. Penelitian ini menunjukan bahwa suplementasi ekstrak daun waru hingga level 100 ppm dapat menurunkan populasi protozoa secara kuadrater dan tidak berpengaruh terhadap peningkatan KBK secara in vitro. Kata Kunci : in vitro, ekstrak daun waru, cairan rumen, populasi protozoa, KBK ABSTRACT The study was aimed to determine the effect of extracted hibiscus leaf on protozoa population and dry matter digestibility in vitro. This experimental research was used randomized block design (RBD), consisted of five blocks, which were the periodical rumen fluid taken from three beef cattles bulls. The treatment tested were the level of extracted hibiscus leaf (Hibiscus tiliaceus) concentrate (mg / kg DM). The level of extracted hibiscus leaf were 0 ppm, 50 ppm, 100 ppm and 150 ppm for RІ, RЇ, R Ј, and RЉ respectively. The ration consisted of 45% ammoniated rice straw and 55% concentrate. The variables measured were protozoa population and dry matter digestibility. The study showed that extracted hibiscus leaf depressed (P <0.05) protozoa population but did not affect (P> 0.05) dry matter digestibility. The increase of extracted hibiscus leaf concentration (ppm) decreased Protozoa population (P <0.05) quadratically with the lowest protozoa population reached at the level of 104,6 ppm. The mean of protozoa population production were 819200 ± 123179.5; 678000 ± 208734.3; 580000 ± 198494.3; 648 000 ± 240977.2 (per ml rumen fluid) and dry matter digestibility were 37.78 ± 2:32; 36.49 ± 2:40; 36.33 ± 2:39; 45.68 ± 19:07 (%) for RІ, RЇ, RЈ, and RЉ respectively. Therefore the supplementation of extracted hibiscus leaf up to 100 ppm could depressed protozoa population, but did not increase the dry matter digestibility in vitro.
