ABSTRACT Aim: The aim of present study was to determine the membrane stabilizing effect of ethanol and aqueous extracts of Humboldtia vahliana Wight on human red blood cell (RBC) membrane. Methods: It was an invitro study, where blood sample was collected from the human volunteers and specific concentrations were made by diluting with buffers. The cell suspensions were exposed to 100 to 1000 μg/mL solution of ethanol and aqueous extracts of Humboldtia vahliana. Cell membrane stability was analyzed with different conditions like change in temperature and pH. Diclofenac sodium was used as standard drug. Each concentration of extracts was performed in a triplicate manner and the data were calculated and presented as Mean with standard deviation. Results: Membrane stabilizing activities were significantly higher with cells which were exposed to ethanol extract of Humboldtia vahliana than aqueousextract. The inhibition of cell membrane rupture was concentration dependent. Conclusion: This study results indicated that both extracts has cell membrane stabilizing activity on human RBC but ethanol extract was potent than aqueous extract. Study report suggested that Humboldtia vahlianabark extracts may offer some beneficial effects in the management of inflammatory mediators. ÖZET Amaç: Bu çalışmanın amacı, Humbildtia vahliana’nın sulu ve etanol ekstraktlarının membran stabilize edici etkisini insan eritrositleri membranı üzerinde değerlendirmektir. Yöntemler: Çalışma invitro koşullarda yapıldı. Gönüllü insanlardan kan alındı ve tamponlarla spesifik dilüsyonlar elde edildi. Hücre süspansiyonları 100 ile 1000 μg/mL arasında Humboldtia vahliana’nın sulu ve etanol ekstraktıyla muamele edildi. pH ve sıcaklık gibi faktörler değiştirilerek hücre membran stabilitesi analiz edildi. Diklofenak sodium standart ilaç olarak kullanıldı. Ekstraktların her bir konsantrasyonuyla deney üç kez tekrarlandı. Sonuçlar ortalama±standart sapma olarak verildi. Bulgular: Etanol ekstraktına maruz kalan hücrelerde membran stabilizasyonu daha fazladır. Hücre membranı rüptüründeki azalma konsantrasyon bağımlı idi. Sonuç: Hem sulu hem de etanol ekstraktının eritrosit membran stabilize edici etkisi bulunmaktadır ancak etanol ekstraktının etkinliği daha fazladır. Humboldtia vahliana kabuk ekstraktı inflamatuvar mediyatörlerin etkinliğinin değiştirilmesinde fayda sağlayabilir.
