Abstract
Populus alba is a huge woody plant. The bark is very white
and smooth when the tree is young. With age, the bark be-
comes darker and more furrowed. This work manipulate the
idea of variaton of the new individual resulted from plant
tssue culture due to any change in media compositon. Pre-
viously it was known that all the new resulted individuals
are genetcally similar, but in this study, the contrast was
proved. As the new individuals are genetcally diferent. The
plant was multplied in shootng and rootng MS media. This
study was designed based on two parameters (media power
and hormone concentratons) resultng in 25 treatments. It
was known that the new plantlets from tssue culture were
identcal, similar to the source mother plant and each other
as well. Nevertheless, the current study manipulated the ef-
fect of various media power and auxins concentratons on P.
alba’s molecular responses. Hence, the genetc stability of
the resulted new individuals from treatments is completely
diferent. Also the diferent auxin hormone concentraton
(IBA and NAA) made genetc instability in the new resulted
individuals. A molecular marker (RAPD-PCR) was used to
estmate the genetc variatons. Four decamers yielded sor-
able bands and led to a total polymorphism percentage of
48.26. The morphological and physiological variaton origi-
nated from the genetc variaton these results confrm that
the new resulted plantlets from diferent tssue culture con-
ditons could be diferent from each other and led to difer-
ent growth parameters.
