145 145 / ﯾﺎﻓﺘﻪ ﻫﺎي ﻧﻮﯾﻦ در ﻋﻠﻮم زﯾﺴﺘﯽ ﺟﻠﺪ7 ، ﺷﻤﺎره2 ، ﺻﻔﺤﺎت145 اﻟﯽ152 ) 1399 ( اﻧﺘﺸﺎرات داﻧﺸﮕﺎه ﺧﻮارزﻣﯽ Nova Biologica Reperta 7(2): 145-152 (2020) Print ISSN: 2423-6330/Online ISSN: 2476-7115 https://nbr.khu.ac.ir; Kharazmi University Press; DOI: 10.29252/nbr.7.2.145 ﺑﺮرﺳﯽ اﺛﺮ ﻧﺎﻧﻮذر ات اﮐﺴﯿﺪ روي ﺳﻨﺘﺰ ﺷﺪه ﺑﻪ روش ﺷﯿﻤﯽ ﺳﺒﺰ ﺑﺎ دﻧﺎ ﻣﻬﻨﺎز ﺟﻨﮕﯽ 1 ، آزاده ﻣﺤﻤﺪ ﻗﻠﯽ 1 و ﻋﺎدﻟﻪ دﯾﻮ ﺳﺎﻻر 2 1 ﮔﺮوه زﯾﺴﺖ ﺷﻨﺎﺳﯽ، داﻧﺸﮑﺪه ﻋﻠﻮم ﭘﺎﯾﻪ، واﺣﺪ ﺗﻬﺮان ﻣﺮﮐﺰي، داﻧﺸﮕﺎه آزاد اﺳﻼﻣﯽ، ﺗﻬﺮان، اﯾﺮان ؛ 2 ﮔﺮوه ﻋﻠﻮم ﺳﻠﻮﻟﯽ و ﻣﻮﻟﮑﻮﻟﯽ ، داﻧﺸﮑﺪه ﻋﻠﻮم زﯾﺴﺘﯽ، داﻧﺸﮕﺎه ﺧﻮارزﻣﯽ، ﺗﻬﺮان، اﯾﺮان ﻣﺴﺌﻮل ﻣﮑﺎﺗﺒﺎت: آزاده ﻣﺤﻤﺪﻗﻠﯽ،a.mohammadgholi@yahoo.com ﭼﮑﯿﺪه. در اﯾﻦ ﻣﻄﺎﻟﻌﻪ از ﻧﺎﻧﻮ ذرات اﮐﺴﯿﺪ روي ﺳﻨﺘﺰﺷﺪه ﺑﺎ ﻋﺼﺎر ه ﭘﻮدر ﻗﻬﻮه ﮐﻪ داراي ﺳﺎزﮔﺎري ﺑﺎ ﻣﺤﯿﻂ زﯾﺴﺖ ﺑﻮده و در اﻧﺠﺎم ﻓﺮ ا ﯾﻨﺪ واﮐﻨﺶ، ﻫﯿﭽ ﮕﻮﻧﻪ ﻣﻮاد ﺳﻤﯽ و ﻣﻀﺮي ﺗﻮﻟﯿﺪ ﻧﻤﯽ ﮐﻨﺪ، اﺳﺘﻔﺎده ﺷﺪه اﺳﺖ . ﺗﻌﺎﻣﻞ ﻧﺎﻧﻮذرات اﮐﺴﯿﺪ روي ﺑﺎDNA ﺗﯿﻤ س ﮔﻮﺳﺎﻟﻪ ﺑﺎ اﺳﺘﻔﺎده از روش ﻫﺎي ﻣﺨﺘﻠﻒ ﻃﯿﻒ ﺳﻨﺠﯽ ﻣﺮﺋﯽ ﻓﺮاﺑﻨﻔﺶ، ﻓﻠﻮرﺳﺎﻧﺲ و ﺗﮑﻨﯿﮏ دورﻧﮓ ﻧﻤﺎﯾﯽ ﺣﻠﻘﻮي ﻣﻮرد ﺑﺮرﺳﯽ ﻗﺮار ﮔﺮﻓﺖ. داده ﻫﺎي ﻃﯿﻒ ﺳﻨﺠﯽ ﻣﺮﺋﯽ ﻓﺮاﺑﻨﻔﺶ ﻧﺸﺎن داد ﮐﻪ ﺑﺎ اﻓﺰاﯾﺶ ﻏﻠﻈﺖ ﺎﻧﻮذرات اﮐﺴﯿﺪ روي، ﺷﺪت ﺟﺬبDNA ﻧﯿﺰ اﻓﺰاﯾﺶ ﻣﯽ ﺎﺑﺪ و اﯾﻦ اﻓﺰاﯾﺶ در ﻧﺎﻧﻮ ذرات اﮐﺴﯿﺪ روي ﺳﻨﺘﺰﺳﺒﺰ در دﻣﺎي اﺗﺎق و ﻓﯿﺰﯾﻮﻟﻮژﯾﮏ اﻧﺠﺎم ﺷﺪ. ﻧﺸﺮ ﻓﻠﻮرﺳﺎﻧﺲ اﺗﯿﺪﯾﻮم ﺑﺮوﻣﺎﯾﺪ ﻧﺸﺎن داد ﮐﻪ ﺑﺎ اﻓﺰاﯾﺶ ﻧﺎﻧ ﻮذره اﮐﺴﯿﺪ روي ﺷﺪت ﻧﺸﺮ ﮐﺎﻫﺶ ﻣﯽ ﯾﺎﺑﺪ ﮐﻪ ﻧﺸﺎن دﻫﻨﺪ ه ورود ﻧﺎﻧﻮ ذره ﺑﻪ ﺳﺎﺧﺘﺎرDNA اﺳﺖ. ﻫﻤﭽﻨﯿﻦ داده ﻫﺎي دورﻧﮓ ﻧﻤﺎﯾﯽ ﺣﻠﻘﻮي ﻧﺸﺎن داد ﮐﻪ اﮐﺴﯿﺪ روي ﺳﻨﺘﺰﺷﺪه ﺑﺎﻋﺚ ﺗﻐﯿﯿﺮ ﺳﺎﺧﺘﺎر ي درDNA ﻣﯽ ﺷﻮد. ﻧﺎﻧﻮ ذره اﮐﺴﯿﺪ روي ﺳﻨﺘﺰ ﺳﺒﺰ ﻣﯽ ﺗﻮاﻧﺪ ﺑﻪDNA ﻣﺘﺼﻞ ﺷ ﺪه و ﺳﺒﺐ اﯾﺠﺎد ﺗﻐﯿﯿﺮ در ﺳﺎﺧﺘﺎر DNA ﺷﻮد ﮐﻪ ﻣﯽ ﺗﻮاﻧﺪ ﺟﺎﯾﮕﺰﯾﻦ ﻧﺎﻧﻮ ذره اﮐﺴﯿﺪ روي ﺷﯿﻤﯿﺎﯾﯽ ﺷﻮد. واژه ﻫﺎي ﮐﻠﯿﺪي. د ي ا ﮐﺴﯽ رﯾﺒﻮﻧﻮﮐﻠﺌﯿﮏ اﺳﯿﺪ، ﯿ ﺳﻨﺠ ﻣﺮﺋ ﻓﺮاﺑﻨﻔﺶ، ﻋﺼﺎر ه ﭘﻮدر ﻗﻬﻮه، ﻏﺪه ﺗﯿﻤﻮس ﮔﻮﺳﺎﻟﻪ، ﻓﻠﻮرﺳﺎﻧﺲ The investigation of the effects of synthesized Zinc oxide nanoparticles on the DNA using green chemistry Mahnaz Jangi 1 , Azadeh Mohammadgholi 1 & Adeleh Divsalar 2 1 Departement of Biology, Faculty of Basic Sciences, Central Tehran Branch, Islamic Azad University, Tehran, Iran; 2 Department of Cell and Molecular Biology, Faculty of Biological Sciences, Kharazmi University, Tehran, Iran Correspondent author: Azadeh Mohammadgholi, a.mohammadgholi@yahoo.com Abstract. In this study, the extract of coffee powder was used to synthesize the zinc oxide nanoparticles due to the compatibility of the method with the environment and the absence of any toxic substance as the byproduct of the reaction. Then, the interaction of zinc oxide nanoparticles with calf thymus DNA was investigated by various spectroscopic methods such as UV-Visible, fluorescence and circular dichroism (CD) techniques. UV-Visible data showed that zinc oxide nanoparticles induced denaturation in DNA in a dose-dependent manner at both the room and physiologic temperatures. The results of extrinsic fluorescence emission of ethidium bromide (EB) also showed that the increase of the zinc oxide nanoparticles concentrations led to the decrease of the emission intensity of EB. This may be the consequence of the intercalation of the nanoparticles into the DNA structure. Also, CD data showed that the synthesized zinc oxide caused structural changes in the DNA. According to the results, it can be concluded that zinc oxide nanoparticles can bind with DNA and induce some structural changes in the DNA structure.
