The compound content in biogas generally consists of methane (CH4), Carbon Dioxide (CO2) and a small amount of Hydrogen Sulfide (H2S). As for nitrogen (N2) and oxygen (O2) usually from the remaining composition of the air analyzed in the sample. The H2S content in biogas is generally more than 1200 ppm which is corrosive so that it often becomes a problem in the surrounding environment because it can damage equipment. One way to minimize H2S compounds is by microbiological processes using microorganisms such as Thiobacillus. Thiobacillus is a chemototrophic bacteria capable of breaking down toxic chemical compounds into non-toxic chemical compounds. Therefore, this research will study the screening process and isolation of indigenous microbes which are able to bind H2S and can be used for biogas purification. This research was conducted in several stages, namely, the process of screening microbes in H2S gas, isolation of potential microbial media containing sulfur NaHS, analyzing data using sulphur analyzer. The results obtained from the stages carried out are the longer the sulfur content will be more and more, on the 20 th day, the sulfur content will increase to 6.201 ppm with a pH of 5. The isolation and screening process for Indonesian microbes (indigeneous) uses conventional isolation methods, namely isolation using the process in stages with isolation stages: the first stage is taking samples from nature, the second stage is diluting the sample in sterile water, the third stage is storing the media for culture, the fourth stage is mixing agar in the petri dish sample, the fifth stage is incubation and the last is the stage of checking the results of the incubation.
