On ISP 2 medium, the yellow-pigmented isolate produced long and branched hyphae that developed into a large number of irregularly shaped spores. It showed growth at 25–30 6C and pH 6.0–9.0 with 0–8 % (w/v) NaCl. Chemotaxonomic and molecular characteristics of the isolate matched those described for members of the genus Promicromonospora. Whole-cell hydrolysates of strain HM 792 T contained the amino acids D- glutamic acid, L-alanine and L-lysine along with the sugars glucose and ribose. The main polar lipids were diphosphatidylglycerol, two unknown phospholipids, two unknown glycolipids and two unknown phosphoglycolipids, complemented by minor concentrations of phosphatidylinositol and phosphatidylglycerol. MK-9(H 4 ) was the predominant menaquinone. The fatty-acid pattern was composed mainly of the saturated branched-chain acids anteiso-C 15 : 0 and iso-C 15 : 0 . 16S rRNA gene sequence analysis showed the highest pairwise sequence identity (96.6–99.0 %) with the members of the genus Promicromonospora. Based on phenotypic and genotypic features, strain HM 792 T is considered to represent a novel species of the genus Promicromonospora, for which the name Promicromonospora iranensis sp. nov. is proposed. Strain HM 792 T (5DSM 45554 T 5UTMC00792 T 5CCUG 63022 T ) is the type strain. The family Promicromonosporaceae Rainey et al. 1997, emend. Zhi et al. 2009 (Stackebrandt et al. , 1997), comprises the type genus Promicromonospora Krasil’nikov et al. 1961 and the genera Cellulosimicrobium Schumann et al. 2001, emend. Brown et al. 2006, emend. Yoon et al. 2007, Isoptericola Stackebrandt et al. 2004, Myceligenerans Cui et al. 2004, emend. Wang et al. 2011, Xiangella Wang et al. 2013, Xylani- bacterium Rivas et al. 2004, Xylanimicrobium Stackebrandt and Schumann 2004 and Xylanimonas Rivas et al. 2003. At the time of writing, the genus Promicromonospora includes ten species with validly published names isolated from soil, sea sediment and air samples, with Promicromonospora citrea Krasil’nikov et al. 1961 as the type species. The members of the genus are characterized by the production of substrate mycelium that fragments into rod-shaped or coccoid elements, the presence of iso- and anteiso-branched cellular fatty acids, MK-9(H 4 ) as the predominant menaquinone and a DNA G+C content of 70–75 mol% (Schumann & Stackebrandt, 2012). In our research programme on finding rare actinomycete species, strain HM 792 T was recognized as a member of a potentially novel species in the genus Promicromonospora. In this study, we determined the taxonomic position of this genomically distinct novel lineage based on a polyphasic approach. Rhizospheric soil samples were collected at ~10 cm depth in Fars Province (Iran) and then first air-dried (Nolan & Cross, 1988), followed by a heat-drying step at 120 u C for 10 min (Nonomura & Ohara, 1971). Strain HM 792 T was isolated by using the dilution plating method on GAC agar (Nonomura & Ohara, 1969) after 14 days of incubation at 28 u C and maintained on yeast extract-malt extract agar [International Streptomyces Project (ISP) 2 medium] slants at 4 u C and as 20 % (v/v) glycerol suspensions at 270 u C.
