Obe- sity is a risk factor for various diseases in- cluding cardiovascular disease, diabetes, & hypertension (Fruh, 2017; Kyrou et al., 2018). Many factors are causing a person to be obese, but mainly due to excessive food intake (Ali & Crowther, 2014). Based on epidemiologi- cal studies, a high-fat diet is closely related to obesity where there is a link between the amount of fat in the food consumed with the degree of obesity (Hruby & Hu, 2015). In the human body, fat degradation is a crucial step in fat metabolism. Fat degradation is carried out by lipase, a key enzyme that plays a role in breaking down triglycerides into mono- acylglycerols and fatty acids in the gastro- intestinal tract (Pirahanchi & Sharma, 2020). In the absence of lipase, fat cannot be absor- bed by the body (Ko et al., 2020). Thus, stra- tegies to prevent and treat obesity can be pur- sued by reducing fat absorption through in- hibition of lipase activity (Birari & Bhutani, 2007; Yun, 2010; Liu et al., 2020). There are many types of lipases, one of them is pancreatic lipases, which are responsi- Citation Aji, O. R., Hudaya, R. A. & Putri, D. A. (2021). In Vitro Pancreatic Lipase Inhibitor Activity of Mangifera foetida Leaves Extract. Jurnal Biodjati, 6(1), 82-92. Received : December 22, 2020 Accepted : May 05, 2021 Abstract. Obesity is a condition where there is excessive fat in the body. Weight loss can be accomplished by inhibiting fat absorption through the inhibition of pancreatic lipase activity, a key enzyme in fat metabolism. Natural products from plants contain various phytochemical compounds that can act as pancreatic lipase inhibitors. This research was conducted to determine the lipase inhibitor activity of the methanol extract of Mangifera foetida leaves. Mangifera foetida leaves were extracted and then the total phenolic and favonoid levels were determined. The extract was tested for its ability to inhibit pancreatic lipase in vitro. The inhibitory activities of the extract and Orlistat were measured using p-nitrophenyl palmitate as a sub- strate at concentrations of 31.25, 62.5, 125, 250, and 500 µg/mL. The results showed that the extracts of young leaves (YL) and mature leaves (ML) contained phenol and favonoid compounds. The IC50 values of young leaf extract (YL) and mature leaves (ML) were 45.22 and 35.50 µg/mL, respectively. In conclusion, the Mangifera foetida leaf extract can be promoted as a good source of anti-obesity com- pounds. Keywords: favonoid, Mangifera foetida, obesity, pancreatic lipase, total phenolic.
