This study uses an in vitro method to determine the nutrition value, antimicrobial efect, and immunomodulatory potential of magot four. The nutrition compound of magot was tested by proximate analytical, sensitivity test was tested by disc difusion method, phagocytosis activity test was observed with peritoneal macrophages of male Balb-C mice (8 weeks old) against Staphylococcus aureus (S. aureus), and challenge of S. aureus against magot four was seen under scanning electron microscopy (SEM). Data of sensitivity test and observation with SEM techniques were reported descriptively. Diferences in phagocytic activity of macrophages between treatments were tested by one-way analysis of variance with honestly signifcant diference (HSD) for further tests. Based on the results we knew that magot four has 38.22% protein with a complete amino acid profle. The highest amino acid content in magot four was glutamate (7,685.84 mg/kg), aspartate (5,864.19 mg/kg), and leucine (5,034.31 mg/kg). The essential fatty acids contained in magot four were lauric acid (13.39%). The sensitivity test results showed that magot four did not sensitive in S. aureus. The introduction of magot four in phagocytosis in vitro could improve the performance of macrophages by its role as opsonin based on SEM observations. The conclusion of this study was magot four could be used as an natural immunomodulator and alternative protein feed source for livestock. Penelitian ini menggunakan metode in vitro untuk mengetahui nilai nutrisi, efek antimikrobial dan potensi immunomodulator tepung magot. Nilai nutrisi magot dianalisa dengan uji proksimat, uji sensitivitas dilakukan dengan metode disc difusi agar, uji aktivitas fagositosis diamati pada makrofag peritoneum mencit Balb-C jantan berumur 8 minggu terhadap Staphylococcus aureus (S. aureus), serta uji tantang S. aureus terhadap tepung magot dilihat di bawah scanning electron microscopy (SEM). Data hasil uji sensitivitas dan pengamatan dengan teknik SEM dilaporkan secara deskriptif. Perbedaan aktivitas fagositosis makrofag antar perlakuan diuji dengan analisis varian satu arah dengan uji lanjut honestly signifcant diference (HSD). Berdasar hasil penelitian diketahui bahwa magot memiliki 38.22% kandungan protein dengan profl asam amino yang lengkap. Kandungan asam amino tertinggi pada tepung magot adalah (7,685.84 mg/kg), aspartat (5,864.19 mg/kg), leusin (5,034.31 mg/kg). Asam lemak esensial yang terkandung pada tepung magot adalah asam laurat (13.39%). Hasil uji sensitivitas diketahui tepung magot tidak sensitif pada bakteri S. aureus. Introduksi tepung magot pada fagositosis secara in vitro dapat meningkatkan kinerja makrofag dengan perannya seperti opsonin berdasar pengamatan SEM. Kesimpulan dari penelitian ini adalah tepung magot potensial digunakan sebagai imunomodulator alami dan pengganti protein pakan unggas.
