Waktu ekuilibrasi sangat menentukan kualitas spermatozoa sapi PO pada proses pembekuan semen. Pada penelitian ini bertujuan untuk mengkaji pengaruh lama ekuilibrasi terhadap viabilitas, motilitas dan abnormalitas spermatozoa sapi PO. Materi yang digunakan adalah seekor sapi PO jantan sebagai sumber semen dan spermatozoa yang dievaluasi. Pada penelitian ini digunakan Rancangan Acak Lengkap (RAL) dengan 4 perlakuan dan 4 kali ulangan. Perlakuan terdiri atas: P1 = Waktu ekuilibrasi selama 2 jam, P2 = Waktu ekuilibrasi selama 3 jam, P3 = Waktu ekuilibrasi selama 4 jam, P4 = Waktu ekuilibrasi selama 5 jam. Variabel yang dievaluasi dalam penelitian ini meliputi kualitas semen (volume, warna, bau, konsistensi dan pH) dan spermatozoa (gerakan massa, konsentrasi, persentase motilitas, viabilitas dan abnormalitas). Data yang diperoleh dianalisis menggunakan analisis ragam (ANOVA) dan dilanjutkan dengan uji Duncan. Hasil penelitian menunjukan bahwa rataan volume sapi PO yang diperoleh adalah 7,63 ml, warna semen putih khas, bau semen khas, konsistensi agak kental-kental, rataan pH 6,3, rataan konsentrasi spermatozoa adalah 1.360 juta/ml dan gerakan massa ++/+++. Sedangkan rataan nilai persentase viabilitas spermatozoa setelah thawing untuk P1(62,63%), P2 (69,50%), P3 (79,63%), P4 (78,25), rataan persentase motilitas untuk P1 (27,50%), P2 (40,00%), P3 (62,50%), P4 (57,50%), dan rataan persentase abnormalitas untuk P1 (2,38%), P2 (2,38%), P3 (1,38%) dan P4 (2,25%). Selanjutnya hasil sidik ragam menunjukan waktu ekuilibrasi berpengaruh nyata (P˂0,05) terhadap viabilitas dan motilotas spermatozoa sapi PO, tetapi tidak berpengaruh nyata terhadap abnormalitas spermatozoa sapi PO. Waktu equilibrasi 4-5 jam menghasilkan angka viabilitas dan motilitas spermatozoa sapi PO yang lebih baik dibandingkan waktu ekuilibrasi 2-3 jam. Kata Kunci : Abnormalitas, Ekuilibrasi, Motilitas, PO, Viabilitas. Abstract. The equilibration determines the spermatozoa quality of Ongole Crossbreed (PO) cattle in spermatozoa freezing procedures. This study aimed to examine the effect of equilibration duration on viability, motility and spermatozoa abnormality of PO cattle. The material used was a bull of PO cattle as a source of semen and evaluated spermatozoa. In this study, a completely randomized design (CRD) was used with 4 treatments and 4 replications. The treatments consisted of four different length of equilibration duration namely 2 hours (P1), 3 hours (P2), 4 hours (P3) and 5 hours (P4). The variables evaluated in this study included semen quality (volume, color, odor, consistency and pH) and spermatozoa (mass movement, concentration, percentage of motility, viability and abnormality). The data obtained were analyzed using analysis of variance (ANOVA) and continued with Duncan's test. The results showed that the average volume of PO cattle obtained was 7.63 ml, the color of the semen was characteristic white, the smell of semen was typical, the consistency was slightly thick, the average pH was 6.3, the average concentration of spermatozoa was 1.360 million/ml and the mass movement was ++/+++. While the average value of the percentage of spermatozoa viability after thawing for P1 (62.63%), P2 (69.50%), P3 (79.63%), P4 (78.25), the average percentage of motility for P1 (27.50% ), P2 (40.00%), P3 (62.50%), P4 (57.50%), and the mean percentage of abnormality for P1 (2.38%), P2 (2.38%), P3 (1 .38%) and P4 (2.25%). Furthermore, the results of variance showed that the equilibration duration had a significant effect (P˂0.05) on the viability and motility of spermatozoa of PO cattle, but had no significant effect on spermatozoa abnormality of PO cattle. The equilibration time of 4-5 hours resulted in better spermatozoa viability and motility of PO cattle than the equilibration time of 2-3 hours.
